I.入侵紅火蟻快速檢測試劑之開發
本分項計畫屬一年期計畫,目的在於應用單元抗體技術開發出"入侵紅火蟻的快速檢測試劑",進一步製成簡易型偵測試紙條棒,提供一般民眾鑑定入侵紅火蟻之用。
II.進口植物檢出重要薊馬分子鑑定技術及偵測鑑定技術之建立及調查
本分項計畫將綜合以上三個年度的研究結果,建立ITS DNA、16S rDNA 及28SrDNA的序列資料庫,建立快速鑑定的基本流程作為協助檢疫工作的依據,除DNA序列的資料庫外並完成PCR-RFLP及Multiplex-PCR的快速鑑定流程。此外將加入數種進口作物常檢出的薊馬,一起分析ITS DNA、16S rDNA 及28S rDNA的資料分子特徵,增加快速鑑定的準確性;包括栗帶薊馬(Anaphothrips sudanensis)、豆花薊馬(Megalurothrips usitatus)、小黃薊馬(Scirtothrips dorsalis)、稻薊馬(Stenchaetothrips biformis)、淡色薊馬(Thrips flavus)以及兩種國內未發現的檢疫薊馬,分別是玫瑰花薊馬(Thri
Abstract: 一.
I.Development of a detecting probe for rapid identification of the red imported fire ant
(Solenopsis invicta)
This project is dedicated to develop a rapid detecting robe for identification of the red imported fire ant (Solenopsis invicta) based on the monoclonal antibody technique. The probe will be further applied to prepare a easy-detecting strip for common people.
II.Molecular identification and monitoring of thrips commonly found in imported crops
The DNA sequences of ITS, 16S rDNA, and 28S rDNA conducted in the past three years will be compiled to establish the identified procedures for quarantine pest. DNA sequences and the derived molecular characters of PCR-RFLP and Multiplex-PCR will provide the identified key characters together. In addition, seven thrips examined commonly from the imported crops will be added in the identified characters this year,including Anaphothrips sudanensis、Megalurothrips usitatus、Scirtothrips
dorsalis、Stenchaetothrips biformis、Thrips flavus and two quarantine thrips of Thrips fuscipennis and Thrips imagines. Finally, the established molecular keys will prove the identified possibility of the unclear thrips found in the imported crops.
III.Development and application of rapid identification techniques for aphids of quarantine importance
The survey aims to develop molecular markers with skills in taxonomy and biochemical/molecular analysis on aphids of quarantine importance with the basis of following results. The program in this year is to develop rapid identification techniques with highly accruacy, readily application and little running time,to standardize current methods and protocols,to produce standard operation procedures to identify aphid pests, with amplification of COI sequences and RFLP protocols,to setup DNA database on aphids of quarantine importance. Finally, as a reference source for quarantine treatment, practical information and relevant scientific evidence with the increasing activity of
international trades by human activities.

The objectives of this research project are to study genetic variation among specialized pathogens using molecular techniques. The long-term goal of this project is to develop the detection techniques for these important pathogens, and provide a standard diagnosis system for the authorities of plant inspection and quarantine. By comparing the host ranges, morphology and DNA sequences, genetic variation of the specialized pathogens will be evaluated. Based on these obtained results, the diagnosis database for specialized pathogens will be established.Furthermore, the specific detection method will be developed for the authorities of plant inspection and quarantine.
