The seeded scaffolds were incubated in culture medium with TGF- beta 3 for 23 days under static or dynamic culture condition . The cell number , amount of matrix, cell morphology and cell differentiation in constructs were determined. The cell number for each scaffold was higher when TGF- beta 3 was added into the culture medium. Cell numbers in CG scaffolds were higher than those in BCII scaffolds. Cell proliferation increased under dynamic culture condition. Many differentiated cells as identified by anti S-100 staining were observed when TGF-beta 3 was employed. The difference in materials of the two scaffolds had not apparent effect of TGF-beta 3 induced hBMSCs transformation into differentiated cells. The dynamic culture system promoted cell proliferation, but not cell differentiation. |
