The uniqueness of this system is: Its thin-film structure can achieve a rapid thermal cycling process; It has a build-in vacuum pouch to generate digital droplet array on-chip. The total distance from the heater to the chamber is 30 μm, and a total of 35 thermal cycles can be completed in 26.5 minutes. To verify the feasibility of this system, miRNA-15b at a range of 125 to 1000 cps/μl are studies. The positive-to-negative ratio of fluorescent intensity of droplets is 10.37, and concentrations of 250 copies and 1000 copies of miRNA-15b can be digitally quantified. Experimental studies also demonstrated that 1000 droplets array can be generated by using the liquid-oil two-phase system. ? 2020 CBMS-0001 |
