摘要:本設施計畫的主要目標為「創建一個世界級的功能性體學研究核心,提供使用者所需的體學服務,藉以厚植台灣生醫實力,提升台灣生技產業競爭力」。在這個計畫中,我們將提供最先進的功能性體學研究平台、一站式單一窗口服務、客製化服務和整合性生物資訊與生物統計服務。藉由此”整合性功能性體學核心實驗室”及其提供的客製化服務,研究同仁將可以完成最尖端的體學研究並達成追求研究卓越的目的。除了先前 NCFPB C5 核心實驗室已提供的服務外,今年,我們將提供與開發下述的新服務項目,第一、基因功能喪失shRNA 基因庫分析晶片。第二、DNA 甲基化定量質譜儀分析。第三、單(少數)細胞基因表現與基因變異分析服務。第四、第二代之新世代定序系統 (HiSeq 和 SOLiD 5500)。第五、第三代之新世代定序系統 (PacificBiosciences)。第六、脂質體分析服務。新世代定序系統和代謝體學是C5 核心重要的服務平台。於今年我們將建立最新的新世代定序系統和代謝體學技術。目前,C5 擁有11套新世代定序系統(六套 SOLiD 4, 一套 SOLiD 5500, 一套Illumina GAII, 一套Illumina HiSeq, 一套Ion Torrent 和一套Pacific Biosciences)可以提供使用者最完整與多樣性的新世代定序服務。SOLiD 5500 和 Illumina HiSeq 具有最大的資料輸出量,IonTorrent 有最快的速度,每次上機時間小於2 小時,Pacific Biosciences NG 可讀取世界上最長的序列,達6000 鹼基對,目前只有C5 提供服務。脂質體學是代謝體學的一個分支,脂質體學是對細胞、組織、生物體內脂質組成與其相互作用的分子進行系統分析的學問,脂質代謝的改變在常見的疾病發病機制中扮演重要角色,是一個快速發展的研究領域。這將是台灣唯一提供分析脂質體學的核心。為了提高服務能力和核心技術,我們將持續開發新的代謝體分析技術,以提供各種樣品的最佳分析效果。我們也將開發新的演算法,以獲得最佳的圖譜分析結果。另一方面,C5 也致力於研發與提供先進與獨特的服務項目。1. 基因功能喪失shRNA 基因庫分析晶片,這是與C6 核心合作提供的服務項目,目前只有C5 提供服務。2. DNA 甲基化定量質譜儀分析,C5 開發DNA 甲基化定量分析,可以作為全表觀基因組分析結果的驗證,此全表觀基因組分析目前全台僅C5 可提供服務。3. 單(少數)細胞基因表現與基因變異分析目前全台也僅為C5 提供的服務,其可於少數細胞或組織中偵測參與重要訊息路徑與性狀的數百個基因。這服務對於瞭解新藥或新治療方式的分子機制極有幫助。在過去的一年間,本核心的服務收入有73,958,230,核心與使用者發表了24 篇論文。C5 核心也申請了3 個專利、參與一個2,000,000 廠商出資的產學計畫,同時培育了34 位碩博士生。我們相信本核心將可以滿足國科會與生技醫藥國家型科技計畫研究人員的迫切服務需求,協助這些研究同仁達成他們最好的表現。
Abstract: The main purpose of this project is to establish a world-class functional omics centerwhich can support the state-of the-art services for researchers to strengthen the basisof medical sciences and the competitive ability of Taiwan biotech industry. In thisproposal, we provide advanced functional omic tools, one-stop service, customized serviceand integrated bioinformatics and biostatistics analysis. By accessing this “Integrated CoreFacility for Functional Omics” and customized services, researchers are able to performcutting edge omic research and achieve research excellence. In addition to previous servicesin NCFPB C5 core facility, in this year we will open new services including 1. Array forloss of function shRNA library screening, 2. MassARRAY for quantitative methylationanalysis, 3. Gene expression and genetic alteration in a single (a few) cell, 4. SecondGeneration NGS service (new generation sequencing)-HiSeq and SOLiD 5500, 5. ThirdGeneration NGS service-Pacific Biosciences, and 6. lipidomics. NGS and metabolomics arethe two important platforms in C5 core facility. We will establish the last NGS andmetabolomics technologies in this year. Right now there are 11 sets of NGS (six SOLiD 4,one SOLiD 5500, one Illumina GAII, one Illumina HiSeq, one Ion Torrent and one PacificBiosciences) providing a comprehensive and diverse NGS for Taiwan researchers. SOLiD5500 and Illumina HiSeq are the highest throughput systems, Ion Torrent is the fastest NGSwith less two hours per run and Pacific Biosciences NGS can measure the longest read up tosix hundred base-pairs in the world only provided by C5 in Taiwan. C5 has become thelargest NGS center in Taiwan. Lipidomics, a branch of metabolomics, is a systems-basedstudy of the entire spectrum of lipids, the molecules with which they interact, and theirfunction within the cell. The alterations in lipid metabolism play important roles in thepathogenesis of most of the common diseases. Lipidomics is a fast growing research field inthe world. C5 is the only core which provides lipidomics analysis in Taiwan. To improve theservice capability and core technology, we will develop analytical method to provide bestperformance for various types of samples. We will also develop new algorithm to obtain thebest data quality for spectrum analysis. On the other hand, we are also devoted to severaladvanced and unique service items: 1. loss of function shRNA library screening service iscollaborated with C6 core facility to serve TRC (The RNAi Consortium) service and onlyprovided by C5, 2. MassARRAY for quantitative methylation analysis is provided tovalidate the DNA methylated loci identified by HumanMethylation450 whole methylationassay only provided by C5, 3. Gene expression and genetic alteration in a single (a few) cellservice is also only provided by C5 and can measured hundreds of gene expressionsinvolved in important pathways and cellular phenotypes that is very helpful forunderstanding the underlying mechanisms of new treatments/drugs. In last year, the serviceincome is NT 73,958,230. Core members and users of this core are published 24 articlesand applied 3 internal patterns. One NT 2,000,000 academic-industry collaboration andformation of two research teams is initiated by C5. We also cultivate 34 Master/PhDstudents. We believe that this core will satisfy the urgent needs from NSC/NRPB researchersand assist the researchers to make their best achievement.
