Abstract
摘要:肝臟為人體中執行代謝、解毒與抗氧化系統之重要器官。但國人肝臟疾病罹患率甚高,如慢性肝病、肝硬化及肝癌等,皆為死亡的主要原因,因此肝臟之保健在台灣是一個重要的課題。此外,近年來已有許多研究證實真菌類(如靈芝和巴西洋菇)與豆科植物(如黑豆和黃耆)中,存有多種機能性成分,能調節人體生理活性。但是,目前仍未有對添加豆科植物作為靈芝或巴西洋菇的發酵基質之研究。
本研究之目的乃在探討「靈芝-豆科」或「巴西洋菇-豆科」發酵產物之抑制肝癌活性與護肝功能,並分析其所含三萜類(triterpenoids)活性成分與抑制肝癌活性之相關性,以及探討抑制肝癌之作用機轉,期能對於「靈芝-豆科」或「巴西洋菇-豆科」發酵產物之機能性能有深入瞭解,以作為健康食品開發之重要參考。本研究擬分三年進行:
第一年:建立快速篩選抑制肝癌活性平台,探討發酵液之抑制肝癌活性及其對大白鼠正常初代肝細胞之影響。此外,進行三萜類化合物之分析,以瞭解三萜類化合物與抑制肝癌活性之相關性。
第二年:藉由四氯化碳(CCl4)誘導大白鼠慢性肝損傷的實驗模式,探討發酵液之護肝功能
Abstract: Liver is the major organ in human being for metabolism, detoxification and antioxidation system. However, the incidence rate of liver diseases is high in Taiwan, such as chronic hepatitis, liver cirrhosis and hepatoma which are the major causes for death. Therefore, how to keep liver health is a big issue here in Taiwan. Recently, many researches have reported that fungi (ex. Ganoderma lucidum and Agaricus blazei) and leguminous plants (ex. Black bean and Astragalus membranaceus) contained many active compounds which can manipulate biological activities. Nevertheless, there are not any researches on the fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants.
The objectives of this project are to investigate the anti-hepatoma activity and liver protective function of the fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants, and to evaluate the relationship between the triterpenoids existing in fermentation broth stated above and anti-hepatoma activity, and also to study the mechanism of anti-hepatoma activity. Hopefully, the physiological functionality on liver protection of the fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants will be elucidated and used as important reference for health food development. This project will be performed in three years:
First year: To establish a high throughput screening system of anti-hepatoma cell lines, and evaluate the effect of fermentation broths with anti-hepatoma activity of G. lucidum and A. blazei cultivated in the medium containing leguminous plants on physiological function of normal primary rat hepatocytes. Then, to analyze the triterpenoids of fermentation broths and to elucidate the relationship between the triterpenoids and anti-hepatoma activity.
Second year: To investigate the effects of various fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants on the carbon tetrachloride-induced chronic liver damage of rats. To evaluate the effects of various fractions of the fermentation broths on the anti-hepatoma activity by high throughput screening system of anti-hepatoma cell lines, and on the physiological function of normal primary rat hepatocytes.
Third year: To investigate the effects of various fractions of fermentation broths of G. lucidum and A. blazei cultivated in the medium containing leguminous plants on the carbon tetrachloride-induced chronic liver damage of rats. Furthermore, the anti-cancer mechanisms of various fractions of fermentation broths on hepatoma cell lines, and the relationship between triterpenoids in fractions and anti-hepatoma activity will also be studied.
