摘要：根據前兩年度對甘藍於高溫淹水、高溫不淹水、適溫淹水及適溫不淹水下之轉錄體，進行定序，找出了各逆境下具差異表現之基因、微型核酸及受影響之代謝路徑。本計畫已完整建立甘藍在高溫淹水逆境下之轉錄體資料庫，了解高溫淹水反應相關之基因以及受調控之微型核酸表現情況。整合前兩年度結果，得知在高溫淹水逆境下共有 2080 個差異表現之基因及 46 個已知微型核酸在此計畫中被找到。本年度預期篩選已知參與代謝路徑及在高溫淹水逆境下有差異表現之基因，利用 Real-time PCR 進行於已知高溫淹水耐受性敏感度不同之甘藍品種個別確認，同時確認生理表現，並建立與高溫淹水相關之轉錄體基因組與生理代謝路徑資料。本計畫之執行將有助於了解甘藍高溫淹水生理及差異表現之基因組，未來或可供做選育高溫淹水耐受品系之分子標記參考使用。
Abstract: According to the results in previous project, trascriptome sequencing of the leaves of cabbage under heat and waterlogging, heat, waterlogging and control was done. The differential expression of genes and the affected metabolic pathways under stresses was studied. Also, we established the database of comprehensive transcriptome of cabbage under heat and waterlogging and understand the expression of genes and micro RNAs involving the responses to these stresses. Based on the transcriptome data, we finally found 2,080 differentially expressed genes and 46 known microRNAs on the stresstreated cabbages. In this year, genes differentially expressed under heat and
waterlogging will be searched and confirmed with Real-time PCR to establish the gene sets and metabolic pathways involving heat and waterlogging. Using cabbage varieties with different stress sensitivity, selected gene sets will be combined with the physiological parameters of cabbages under stress to demonstrate the relation between genes and stress tolerance. The execution of this project will be helpful to understand the physiology of heat and waterlogging of cabbage. The differentially expressed gene sets and involved micro RNA might be possible molecular makers for selecting heat and waterlogging tolerant cabbage lines.
