Abstract
摘要:我們與西安交通大學研究團隊合作,研究目標在於利用生物耦聯金奈米環及奈米柱之侷域表面電漿子共振所產生螢光加強、散射提昇、單態氧產生率提高以及吸收強化等效應來達到結合光學同調斷層掃瞄與螢光影像之癌細胞標記應用和結合標靶光熱療法與標靶光動力療法之癌細胞滅活應用。基於個別之研究專長,臺灣大學團隊將進行生物耦聯金奈米環之製作,環之尺寸小於100奈米以利於癌細胞之吞噬,其侷域表面電漿子共振波長大於950奈米以利光線能更深透入人體組織。侷域表面電漿子共振所產生之同調散射增強配合干涉本質之光學同調斷層掃瞄可用來瞭解細胞吞噬金奈米環之行為,並達到癌細胞標記的效果。同時侷域表面電漿子共振所帶來的吸收增強可用於光熱效應之產生,以達到癌細胞滅活的效果。另一方面,西安交通大學團隊將合成生物耦聯金奈米柱來產生侷域表面電漿子共振,以改善螢光影像,達到癌細胞標記的效果,同時提高光敏劑之單態氧產量,以加強光動力療法作用,達到癌細胞滅活的效果。本兩岸合作計畫之研究重點在於結合光學同調斷層掃瞄及螢光影像的癌細胞標記技術和結合光熱療法與光動力療法的癌細胞滅活技術。這種結合之技術可望為將來之腫瘤診斷與治療提供一個有效的多功能光學方法。
Abstract: In cooperating with a research team of Xi`an Jiaotong University (XJTU), this joint research project aims at the cancer cell labeling with the combination of optical coherence tomography and fluorescence imaging and the cancer cell inactivation with the combination of targeted photothermal therapy and photodynamic therapy using the enhanced emission, scattering, singlet-oxygen generation, and absorption of localized surface plasmon (LSP) resonance of bio-conjugated Au nanorings and nanorods. Based on their individual expertise, the National Taiwan University (NTU) team will be working on the fabrication of bio-conjugated Au nanorings of <100 nm in size for inducing LSP resonance of >950 nm in resonance wavelength. The small Au nanoring size is designed for effective uptake by cancer cells. The long LSP resonance wavelength is required for the future application of deeper tissue penetration. The LSP-resonance enhanced coherent scattering of Au nanoring will be monitored with interferometric optical coherence tomography for understanding the Au nanoring uptake behavior by cancer cells and implementing cancer cell labeling. Also, the LSP-resonance enhanced absorption of Au nanoring will be used to generate photothermal effect for cancer cell inactivation. On the other hand, the XJTU team will synthesize bio-conjugated Au nanorods to induce LSP resonance for improving fluorescence imaging (for cancer cell labeling) and enhancing the singlet-oxygen generation of the linked photosensitizer in photodynamic therapy (for cancer cell inactivation). The key issues of the cross-strait research cooperation include the combination of the cancer cell labeling techniques of optical coherence tomography and fluorescence imaging, and the combination of the cancer cell inactivation techniques of photothermal therapy and photodynamic therapy. Such combinations will lead to a multi-functional optical modality for tumor diagnosis and therapeutics in the future development.
