本研究計畫探討SIRT5對於骨細胞抵禦缺氧壓力的機制,可能來自其調控硫鍵傳遞系統來防止橋接蛋白p66shc形成四聚體進入粒線體,產生ROS造成細胞刁亡。我們可以運用提高SIRT5活性的藥物1,4-dihydropyridine,來降低p66shc的活化,緩解缺氧導致的成骨細胞刁亡與骨質破壞。對於牙醫來說,防止受傷的齒槽骨因缺氧而壞死,加速齒槽骨在手術後的癒合,或是防止電療、藥物造成的骨壞死等等,都是重要的議題。我們的研究在探討骨細胞面對缺氧及氧化壓力的機制,並找尋可以抑制缺氧傷害的方法。
