視神經病變會造成視網膜神經節細胞受損,進而導致視神經萎縮和視力受損。衣康酸是檸檬酸循環的中間產物,具有抗發炎、抗氧化、調節免疫功能的角色,其臨床應用近年來引起許多關注,但在視神經病變治療上並沒有相關研究報告。本研究首先以初代培養之小鼠視網膜神經節細胞模型,觀察4-辛基衣康酸(4-OI)對細胞凋亡的保護作用,並釐清其作用機轉。接著在動物實驗中,藉由經鼻給藥模式,以質譜分析眼睛內衣康酸濃度,確認其藥物動力學。然後選擇視神經壓傷和實驗性自體免疫腦脊髓炎小鼠模型,驗證經鼻給予4-OI對神經細胞的保護作用,確認視覺功能的變化、視神經細胞存活率,並且釐清其訊息傳遞作用機轉。希望提供視神經病變治療的新方向。
