我們利用CRISPR/Cas9基因編輯系統,於全球首次證明其能在不造成雙股DNA斷裂下,將HBV integrated DNA 與cccDNA永久去活化。本計畫更進一步利用於Streptococcus canis發現的scCas9與最新prime editing工具,建立CRISPR/Cas9衍伸出之鹼基編輯(base editing)工具,針對HBV啟動子與open reading frame,找尋適用於不同HBV基因型的保守標的序列。也將利用split- Cas9技術,改善AAV載體之小鼠動物模式體內傳輸效率,檢測其治療B型肝炎之可行性。此研究成果有潛力發展出功能性根除B肝治療策略。
