Abstract
Plant cells possess totipotency that
means whole plants could be
regenerated from single cell by
modulating culture conditions. The
mechanisms of totipotency, however,
were limitedly understood so far.
From thorough studies, it had found the
contents of endogenous IAA and ABA
were increased significantly in rice
callus by osmotic stress treatment, and
were correlated to the ability of shoot
regeneration. We, therefore, did
further studies to explore the correlation
among phytohormones, carbohydrate
metabolism and osmotic stress during
shoot regeneration.
TN5-AnA callus was induced on MSD10 basal medium containing
anthranilic acid and ABA. The results
showed all the phenomenons were
similar to those under osmotic stress
treatment. Higher IAA and ABA levels
inhibited all the sucrolytic enzyme (Sol-
IT, Bound-IT, and SS) and amylase
activity. Therefore, the higher soluble
sugars and starch contents might be
resulted from sucrose uptaking by
sucrose transporter dominantly and
inhibiting starch degradation,
respectively. In conclusion, osmotic
stress induced some changes of
carbohydrate metabolism, and was
brought about shoot regeneration. Parts
of those changes were regulated by
changes of endogenous IAA and ABA
levels. But there are some processes
induced by osmotic stress might be
regulated by other factors during shoot
regeneration.
