Abstract
This project was conducted to
overexpress E.coli and ruminal phytase
genes in rice plants by using
Agrobacterium-mediated transformation
approach. We get 120 independent
putative transgenic rice plants(T0)from
8 different plasmid constructions.
Southern blot analysis showed that most
of the transformants harboring 1 to 4
copies of transgene in rice genome.
Northern blot analysis revealed that
maximum phytase RNA expression can
be obtained in seeds after 4-5 days
germination. Besides, enzyme activity
assay confirm that higher RNA
expression also possess higher phytase
activity. Moreover, phytase activity of
transgenic plants is hundred folds than
that of nontransformed one. Agronomic
characrestics of T1 plants will be
analyzed further.
