Abstract
Limb allotransplantation is a combination of composite tissues including a great number of highly antigenic and immunocompetent tissues. In animal model study, limb grafts usually encounter uncontrollable rejection and other complications including graft-versus-host
disease (GVHD). In our recent studies the rejection of limb grafts may be successfully controlled by combination of novel immuno suppression agents with less side- effects. In this study, limb composite tissue allotransplantation is carried out with a combination of immunosuppressants , bone marrow transplantation, and chemo T cell depletion agents. As a result, this model did not develop a reliable GVHD model, however, some animals developed tolerance after stopping immunosuppressants when the agents were with enough dosage. Under the condition of underdose immunosuppression, no microchimerism was accomplished, and the bone marrow cells may lead to early rejection even GVHD. A further refined immunosuppression protocol is mandatory in future composite tissue trnasplantation.
