Abstract
The investigation of nutrient storage, floral quality and floral time in Oncidium Gower
Ramsey pseudobulb during the development of flower stalk were carried out. At the
initiation of flower stalk (length 2-4 cM), more than 1000 pseudobulb specific expressed
genes were discovered using cDNA microarray and subtractive EST library; The Northern
blot analysis of fourteen cDNA in the subtractive EST library related to carbohydrate
metabolism group confirm that thirteen cDNA have higher expression level in pseudobulb,
suggesting mucilage may be formed first and then transferred to starch during the
development of flower stalk. We also found that class III peroxidase (POXs) were highly
expressed in the subtractive EST library and obtained eleven POXs genes form Oncidium
cDNA library. Now these POXs genes have been transformed to Arabidopsis and
Oncidium plants to observe their effects on inflorescence and flower development.
