Comparison of gene expression profile and development of red spotted grouper (Epinephelus coioides) larvae fed with copepod and Artemia

Abstract
Grouper is important culture fish with high economic value in Taiwan, the major living feed during the middle stage of grouper fry culture are artemia and Copepods. To understand the effect of different living feed on fish fry development, we first test the anti-stress survival rate that remove from water of grouper fry fed with different living feed, the survival rate of the group fed with artemia was 8.13%, the group fed with artemia enriched with super SELCO was 72.28%, and the group fed with Copepods was 98.21%. Then we further investigated the fatty acid composition of living feed and grouper. In the living feed, artemia contains DHA 0.22%, EPA 1.80%, ARA 0.48%; artemia enriched with super SELCO contains DHA 4.53%, EPA 5.12%, ARA 0.66%; Copepods contain DHA 6.30%, EPA 8.48%, ARA 1.67%. And in fish fry, the group fed with artemia contains DHA 1.66%, EPA 5.24%, ARA 3.28%, fed with artemia enriched with super SELCO contains DHA 7.14%, EPA 6.44%, ARA 3.81%, and the group fed with Copepods contains DHA 15.47%, EPA 7.13%, ARA 1.57%. In which, only the group fed with Copepods completes fry metamorphosis after 14 days of feeding period. The results showed that the content of highly unsaturated fatty acid in food chain has significant influence on the development, metamorphosis and anti-stress survival rate of grouper fry. We also used microarray to analyze the differential gene expression of fry fed with artemia and copepods, and found that there were 253 genes with up-regulated and 123 genes with down-regulated. These differentially expressed genes include glycosylation related genes: muscle-beta-nolase 3, pyruvate kinase, muscle-phosphoglycerate mutase 2-1, glyceraldehyde-3-phosphate dehydrogenase and fructose-bisphosphate aldolase A; nucleic acid synthesis and cellular regulatory related genes: nucleoside diphosphate kinase B and nucleoside diphosphate kinase-Z1; structural gene:lily-type, lectinmuscle-specific beta 1 integrin binding protein 2 and similar to Histone H1.5.
