Abstract
Multifunctional Drug Delivery Systems with both diagnostic and therapeutic capabilities are desired in recent researches. In this study, we design and synthesize a wormlike mesoporous silica (WMSN) based drug delivery vehicle that combines extremely small iron oxide nanoparticles (ESIONPs) on the outer mesoporous silica shell, controlled release using boronic acid functionalized gold nanoparticles as nanoscopic caps and targeting therapy with the help of folic acid (FA). In diagnoses, T1 MRI images from ESIONPs and Computer Tomography (CT) images from AuNPs provide precise information while in therapies, controlled release and targeting offer accurate release of drug to the destinations which increase the specificities and reduce side effects. To the best of our knowledge, this is the first time that encapsulate T1 signal ESIONPs inside the network of mesoporous silica nanoparticles acting as a nanocarrier to perform MRI/CT dual imaging, controlled release and targeting therpy. Besides, the boronate ester bonds which connect AuNPs and WMSN are reversible and can be hydrolyzed via the change of pH values or in the presence of cis-diols which enhances its applicabilities under different physiological conditions. Furthermore, the targeting ligand, folic acid, can also be displaced with other targeting molecules and thus increases the selectivities of the drug delivery system to other cell lines. We expect its contributions to the studies of biomedicines of the future as a mature model of drug delivery system with both diagnostic and therapeutic capabilities. It is actually a miniature aircraft carrier.
