Abstract
This chapter is a review of clinical trial and observational data on cardiovascular and upper gastrointestinal safety of nonsteroidal anti-inflammatory drugs, with a focus on the risks associated with selective cyclooxygenase-2 inhibitors. The review covers the safety signal identification, signal refinement, and signal evaluation for rofecoxib and celecoxib from 2000 through the latest publications. Implications for the future of science of safety for medical products are discussed.
