Abstract
There are two major nonparametric methods for the test of linkage.One is the mean test which is applied to affected sib pairs.The other is TDT
(transmission/disequilibrium test)which is applied to case-parent triad families and can be extended to multiple affected children.Therefore,when the data are gathered with affected sib pairs,both methods are applicable for the test of linkage.It has been known that when linkage disequilibrium is weak,the power of mean test is greater than that of TDT,but when linkage disequilibrium is strong,the power of TDT is greater than that of the mean test.In this paper,we propose a new method to combine two tests to construct a new statistic. First,we derived their joint probabilities for the transmission-nontransmission information and identical by descent information in sib pairs.We showed that the two variables representing the two types of information are dependent.Based on this joint probabilities we developed a new statistic for the test of linkage.Finally,we compared the powers of the new statistic,TDT and the mean test.
