Abstract
Duck viral hepatitis (DVH) usually
affects young ducklings and caused high
mortality. The disease is characterized with
typical hepatitis lesions. In Taiwan, two
major outbreaks of DVH occurred in 1972
and 1990 respectively. Up to date, no
nucleotide sequence f DVH virus has been
revealed. So the study on the virus are rare.
In this project, we retrieved the viral
nucleotide sequence of the Picornaviridae
family from Genebank of NCBI. Then we
used the cluster method of Megalign
program in Clustal WR and DnastarR 分
package to conduct the multiple
sequence comparison. Based on the
results of the comparison, degenerate
primers were designed according to the
different virus genus in the
Picornaviridae and targeting to the
different location. After conducting
RT-PCR, one set of the primer could
generate a 589bp products. The RT-PCR
product was sequenced and phylogenetic
analyzed by DnastarR及PhylipRprograms.
The results indicated that DVH virus is
more related to the human rhinovirus
and porcine enterovirus.
