Endogenous ornithine and arginine contents and dark-induced proline accumulation in detached rice leaves

Abstract
Since proline accumulates in detached rice leaves under dark condition, we determined the mechanism of this dark-induced process. In darkness, proline content in detached rice leaves incubated in distilled water did not increase at 12 h after incubation but increased about 2- and 2.5-fold at 24 and 48 h, respectively. Proline content, however, in detached rice leaves incubated in the light remained relatively unchanged in 48 h. In contrast, proline accumulation in detached rice leaves incubated in ornithine and arginine but not glutamic acid at the concentration higher than 10 mmol·L-1 was greater in the light than in darkness. We also observed that endogenous ornithine and arginine contents in detached rice leaves in the light were much lower than those in darkness. All these results suggest that high contents of ornithine and arginine are associated with dark-induced proline accumulation in detached rice leaves. There was no difference in the activity of ornithine-δ-aminotransferase, an enzyme responsible for proline accumulation via ornithine pathway, between light- and dark-treated detached rice leaves.
