Antrodia xantha，Fomitopsis pinicola，和 Rigidoporus ulmarius之成份分析及抗發炎之生物活性研究

Abstract
開發優質高產的藥用真菌，是本研究的目標。本論文選用3個藥用真菌品種，分別是Antrodia xantha，Fomitopsis pinicola （Swartz. Fr.） Karst.，Rigidoporus ulmarius（sow.:Fr） Imaz. （選用3個品系，分別是TFRI 1047，MG TFRI 1047，MG TFRI 1058），在培養35天後，其中在菌絲體乾重的產率方面，MG TFRI 1058可以達到15.17 g/l的產量；在多醣體產量方面，TFRI 1047可以達到197 mg/l的產量；在乙醇萃取物方面，MG TFRI 1058可以達到6.2 mg/l的產量。在生物活性方面，MTT細胞毒性結果顯示，在1000 μg/ml 的濃度下，以上5個菌種的菌絲多醣體及乙醇萃取物都不具有生物毒性。於外加interferon-γ於人類靜脈內皮細胞 (human vascular endothelial cell, ECV cell)，以引起細胞之發炎反應，及促使細胞中IP-10蛋白之表現後，再加入菌絲萃取物後觀察其抑制IP-10蛋白及其啟動子表現的情形，以測定是否有抗發炎的能力。結果顯示，在多醣體方面，Antrodia xantha，Rigidoporus ulmarius都具有抑制IP-10蛋白質釋出的能力，其中又以Rigidoporus ulmarius中的MG TFRI 1047，在1 μg/ml 的濃度處理下，降低67 %的IP-10蛋白質釋出為最佳；在乙醇萃取物方面，Antrodia xantha，Fomitopsis pinicola，Rigidoporus ulmarius中品系MG TFRI 1047和MG TFRI 1058都具有抑制IP-10蛋白質釋出的能力，其中又以Fomitopsis pinicola，在100 μg/ml 的濃度處理下，可以降低58 %的IP-10蛋白質表現，及MG TFRI 1058，在100 μg/ml 的濃度處理下，可以降低90 %的IP-10蛋白質表現具有較好的表現。
在本論文的結論中，我認為多醣在抗發炎上的能力上是所有萃取物中較好的，其中又以Rigidoporus ulmarius中的MG TFRI 1047是未來最有發展性的菌種，因為此菌種不論是在產量上或是抗發炎活性上都具有最好的表現。
