Abstract
In the dot blot analysis with total
RNA of lily leaves treated with salicylic
acid (SA), the barley PR-4, 14-3-3,
chalcone synthase and oat tlp gene
probes detected stronger signals,
indicating that related genes may present
in lily and are SA inducible. In order to
perform northern hybridization analysis,
the 18S rDNA of lily was cloned and
sequenced after PCR amplification. In
order to clone the cDNA fragments of
defense genes of lily in response to the
induction of chemicals, RT-PCR was
perfomed using primers designed
according to the sequences of chitinase,
barley PR-4, and oat tlp genes. A cDNA
fragment with similar size to the
prediction was amplified. The amino
acid sequence showed 32% similarity to
2
that of cysteine proteinase of tomato, a
protein expressed in cold temperature.
This cDNA fragment was presumed
related to the defense reaction of lily.
