Abstract
Surface modification by polyelectrolyte layer-by-layer (LBL) deposition allows coating substrates with complicated geometries and is extremely versatile and useful for biomedical devices. Several parameters in preparation of polyelectrolyte multilayer (PEM) films, such as the number of layers and the assembly conditions, might influence the interactions between the surface and biological system. The aim of this study is to investigate the influence of number of layers of PEM films on the behavior of osteoblast-like cell line, MG-63. Furthermore, RGD was used to modify the PEM and the modulation of cell functions on the RGD-modified PEM was studied. Poly(acrylic acid) (PAA) and Poly(allylamine hydrochloride) (PAH) solution were assembled onto TCPS by LBL technique with different layer numbers and different topmost layers. MG-63 was seeded onto the PEM film-coated plates, and cell adhesion, growth and alkaline phosphatase (ALP) activity were studied. We found that cell adhered and proliferated better on the surfaces with fewer layers. Moreover, the cells’ ALP activities decreased with the increasing number of layers. Cells spread on all the surfaces on Day 1, while cells aggregated on the surfaces with more than 5-layer PEM films after 3 days, but not on TCPS and 4-layer surface. Our results indicate that different layer numbers influence cell behavior. However the charges of the top most layer was not found to affect the cell behavior. RGD functionalized PEM enhanced cell adhesion, proliferation and differentiation. Cells adhered better on the PEM film which was built in pH6.5 than pH2.0, but cells stopped proliferation on the both films after day 3. Cells on RGD functionalized PEM were not form aggregates after day 3. On the other hand, cells proliferated and differentiated faster on 6.5-RGD compare with2.0-RGD. Our result showed that cell behavior is dependent upon the pH of PEM built up solution . These works support the potential of PEM films for surface modification of biomaterial, and point to a novel prespective for peptide functionalizing that can lead to an improvement of biocompatibility.
