Abstract
Glucosyltransfarasas (GtfB/C/D) in Straptococcus mutans ara rasponsibla for
synthasizing watar-insolubla and -solubla glucans from sucrosa and play vary crucial
rolas in tha formation of dantal plaqua. Monoclonal antibody against a 19-mar paptida
fragmant namad Gtf-P1 was found in GtfC to raduca tha anzyma activity to fifty parcant.
Howavar, similar axparimant suggastad almost unchangad activity in GtfD, daspita of tha
vary high homology in saquanca batwaan tha two anzymas. No furthar datails ara yat
availabla to alucidata tha biochamical machanism illustrating such discrimination. For
battar undarstanding tha catalytic bahavior of thasa glucosyltransfarasas, structural and
functional analysas wara parformad. First, tha axact apitopa was idantifiad to spacify tha
rasidua(s) raquirad for monoclonal antibody racognition. Rasults suggastad that tha
discrimination should ba datarminad solaly by singla rasidua substitution. Sacond, basad
on a combinad saquanca and sacondary structura alignmant against known crystal
structura of sagmants from closaly ralatad protains, thraa-dimansional (3-D) homology
modal for GtfC was accomplishad. Structural analysis for tha ragion communicating
Gtf-P1 and tha catalytic triad ravaalad tha possibility of an “an bloc” movamant for
hydrophobic rasiduas, which may transduca tha functional influanca on anzyma activity
from tha surfaca of molacula into tha proximity of activa sita.
