Abstract
The mZFG-1 gene (mouse zinc finger gene-1; Gene bank accession number:
AF204776) that was first identified from early mouse embryos using the DNA
sequencing shotgun strategy coupled with in silico mining of mouse EST database.
Using the mouse EST sequence-based primer sets for RT-PCR, the mZFG-1 mRNA
was confirmed to be expressed in mouse pre-implantation embryo and in reproductive
organs of adult mice including uterus, ovary and testis. By further library screening
using this cDNA fragment as a probe and 5’-RACE techniques, two (short and long)

forms of the mZFG-1cDNAs were obtained. Since both mRNAs encode proteins with
interspersed zinc finger motifs, we refer to the short and long mZFG-1 transcripts as
mZFG-1S and mZFG-1L, respectively. RT-PCR and Northern blotting nanlyses
demonstrated the mZFG-1L mRNA was expressed only in adult testis. The short form
cDNA was ubiquitously found in male and female organs and early pre-implantation
embryos. Localization of these transcripts using in situ hybridization further
confirmed that expression of mZFG-1L occurred only in Sertoli cells; However,
mRNA of mZFG-1S was found in Sertoli and Leydig’s cells of testis and in Granulosa
cells of ovary.
