Abstract
This project attempted to investigate the
toxic effect of nitrogen oxides (NOx) and
polycyclic aromatic hydrocarbons (PAHs).
They are environmental pollutants commonly
found in the exhaust released from
imcomplete combustion. In addition, NOx
may attack PAHs to form potent mutagenic
nitro derivatives. First year, we investigated
combined effects of NOx and PAHs in
animal models. We found that the
mutagenicity of combined exposure was
higher than that of pyrene exposure alone.
Second year, we measured the levels of
plasma NOx and urinary 1-hydroxypyrene
(1-OHP) for the taxi drivers and the referent
groups. We recruited ninety-five male taxi
drivers and 75 male community residents as
the referents. With consent, subjects provided
blood and urine samples and completed a
self-reported questionnaire. Significant
difference in the average NOx level was
observed between drivers and referents (45.6
vs. 40.0 μM). The drivers had higher level of
1-hydroxypyrene (1-OHP) compared with
the referents (0.33 vs. 0.20 μg/g creatinine).
We used urinary 8-hydroxydeoxyguanosine
(8-OHdG) as an effective biomarker on DNA
damage due to NOx and PAHs exposure
from traffic exhaust, and susceptible gene
MspI/HincII and GST M1/T1 polymorphism
related to exposure level and DNA oxidative
damage. In the multiple linear regression,
after adjusting for smoking, GSTM1
genotype and other associate variables, there
was a significant correlation between the
levels of urinary 8-OHdG and 1-OHP but not
NOx. This study supports that taxi drivers
had higher levels of NOx and 1-OHP due to traffic exhaust exposure and 8-OHdG level
may serve as a sensitive biomarker of induce
oxidative damage associated with PAHs
exposure.
