Abstract
This research has successfully used all the technology to design and produce a biological chip. It reconstructs the experiment procedure of the Progesterone test kit used by Department of Animal Science and Techonolgy, NTU, which reduces the experiment time and dose. After the experiment dose is given, the chip can achieve the same result in general Enzyme-immunoassay by using a designed way, which allows the dose to enter the reaction zone under the control of the peristalsis pump, and by using the Spectrophotometer to figure out the concentration of the dose.
After the experiment, we decided to use Polydimethylsiloxane (PDMS) as the material to make the chip. Each chip has five passages, respectively carrying different buffer solution. With different lengths of the passages, the Enzyme-immunoassay can occur in the chip after the absorption of the antibodies has been verified.
Finally, by successfully integrating biological chip technology with Enzyme-immunoassay, this research has produced a biochip to replace the traditional, time-consuming experiment. This chip is time-saving, low-cost, highly sensitive and selective. It provides the future dairy farmers who wish to monitor their cows during the rut with a portable, convenient and stable tool.
