Abstract
Recently, there is increasing public and
scientific concern that some environmental
pollutants may cause hormone disruption in
human and wildlife. Therefore, there is an
urgent need to develop an effective assay
which can detect environmental hormone. In
this study, an integrated bioassay which
using acute responses of prepubertal female
rats will be expanded and developed to
examine effects of PCB 132 (2,2’,3,3’,4,6’-
hexachlorobiphenyl) and PCB 149
(2,2’,3,4’5’6-hexachlorobiphenyl) on
estrogenicity, hepatic enzyme activities,
thyroxine, and brain dopamine as well as
norepinephrine levels. First year of this
project, it was focused to standardize the
methods of the uterotropic responses as well
as hepatic enzyme activities. The result of
this study showed that PCB 132 and PCB
149 did not produce any uterotropic
responses in prepubertal female rats at 8
mg/kg, 32 mg/kg and 96 mg/kg. PCB 132
and PCB 149 did not increase liver
microsomal P450 1A enzyme activities of
prepubertal female rats. However, there was
an increase P450 2B activity at prepubertal
female rats administered PCB 149 at 96
mg/kg. It indicated that PCB 149 was a PBtype
enzyme inducer. The toxic effects of
2,3,6-substitiued PCB congeners were nondioxin-
like.
