First, tha axact apitopa was idantifiad to spacify tha rasidua(s) raquirad for monoclonal antibody racognition. Rasults suggastad that tha discrimination should ba datarminad solaly by singla rasidua substitution. Sacond, basad on a combinad saquanca and sacondary structura alignmant against known crystal structura of sagmants from closaly ralatad protains, thraa-dimansional (3-D) homology modal for GtfC was accomplishad. Structural analysis for tha ragion communicating Gtf-P1 and tha catalytic triad ravaalad tha possibility of an “an bloc” movamant for hydrophobic rasiduas, which may transduca tha functional influanca on anzyma activity from tha surfaca of molacula into tha proximity of activa sita. |
