標題: | 行政院國家科學委員會專題研究計畫成果報告:免疫注射與隨後以弓蟲攻擊對細胞反應及細胞素表現之影響 | 作者: | 林大盛 | 關鍵字: | T 細胞;細胞素;細胞免疫;弓蟲;T cell;cytokine;cell-mediated immunity;Toxoplasma gondii | 公開日期: | 1998 | 出版社: | 臺北市:國立臺灣大學獸醫學系暨研究所 | 摘要: | 在弓蟲抵抗力方面,細胞免疫(CMI, cell mediated immunity) 比抗體重要。CMI 之作用,主要藉T cells 分泌各種cytokines 進行。一為Th1-cytokines 如IL-2 及IFN-g; 一為Th2-cytokines 如IL-4,IL-6,及IL-10, 二者互相拮抗。一般而言,Th1-cytokines 有抑制弓蟲效果,而Th2-cytokines 卻促使 疾病嚴重化。IFN-g可活化macrophages 以 消滅弓蟲。另外,IL-6 有助於細胞內弓蟲 繁殖。而IL-10 可抑制被IFN-g活化之 macrophages 。然而最近發現,Th1- 與 Th2-cytokines 的關係並非兩極化。如IL-4 在感染初期, 因抑制proinflammatory cytokines 產生,反而減低小鼠感染弓蟲之 死亡率。IL-10 亦發現可抑制因 proinflammatory cytokines 造成之弓蟲性腦 部病變。另外,雖然常同時表現兩型的 cytokines,但仍以其中一型為主。故茲欲以 弓蟲強毒株RH,探討小鼠免疫注射與隨後 弓蟲攻擊,來自不同性別之細胞對弓蟲抗 原及mitogen 之反應,與其cytokines 分泌 的變化,藉以了解各種cytokines 扮演的角 色與相互關係。 It is evident that cell-mediated immunity (CMI) is more important than antibody in protective immunity against Toxoplasma gondii. The effect of CMI is mediated by cytokines released mainly by T cells. Th1- cytokines include IL-2 and IFN-g; and Th2- cytokines include IL-4, IL-6, and IL-10. They are antagonists. In general, Th1- cytokines inhibit T. gondii proliferation and Th2-cytokines promote severity of the disease. IFN-g activates macrophages that kill T. gondii. In addition, IL-6 assists intracellular proliferation of T. gondii, and IL-10 can inhibit the activity of IFN-g- activated macrophages. However, recent findings indicate that the relations between Th1- and Th2-cytokines may be more complicated. In the early stage of infection, due to the inhibition of proinflammatory cytokines by IL-4, the mortality of infected mice is reduced. Similarly, IL-10 can restrain toxoplasmic pathology in brain caused by proinflammatory cytokines. Although both types of cytokines may be released, one type will be dominant. In the present study, virulent strain RH will be used to study antigenic and mitogenic responses of cells from different sex, and the changes of cytokine production in mice after immunization and subsequent challenge with T. gondii, in order to realize the roles played by various cytokines. |
