標題: | 家禽腺病毒纖毛基因與毒力關係之研究 | 作者: | 蔡向榮 | 關鍵字: | 家禽腺病毒;纖毛基因序列;聚合脢連鎖反應;Fowl Adenovirus;Fiber Gene Sequence;Polymerase Chain Reaction | 公開日期: | 2001 | 出版社: | 臺北市:國立臺灣大學獸醫學系暨研究所 | 摘要: | 以家禽腺病毒(FAV) 之CELO 株 penton 之核酸序列所設計之1 對聚合脢連 鎖反應引子,可快速偵測所有11 株台灣分 離株(TFAV)。依據澳洲分離株CFA40 之纖 毛基因序列設計之1 對引子,進行纖毛基 因之PCR-RFLP 及序列分析比較9 株 TFAVs 與2 株(CFA3 和CFA40)澳洲罹 患腺病毒包涵體肝炎分離之FAV (AFAV) 及2 株(FAV 1A 和FAV 10)標準株FAV (SFAVs) ,結果TFAV 可被分成2 (I 和II) 大群。雞源TFAV 與2 株AFAVs 同屬第I 群中之第一小群(I-1),而2 株低病原性之 雞源SFAVs 則屬於第I 大群中之第二小群 (I-2)。鴿源及鴨源TFAVs 在纖毛基因之岐 異度大,與產蛋下降症病毒(EDSV) 亦有 某種程度上之差異,但與EDSV 同屬第II 群。此結果顯示FAV纖毛基因可能與FAVs 對各種動物之病原性有關聯。 We designed a pair of primers based on the sequence of penton of CELO fowl adenovirus (FAV) for polymerase chain reaction (PCR) and found that the PCR can detect all 11 Taiwan strains. Another pair of primers was also designed based on the fiber gene sequence. Utilizing the primers combined with the restriction enzyme profile, the product of PCR from 9 of 11 Taiwan strains were sequenced and compared with each other and the published fiber sequence of CFA3, CFA40, FAV-1A, FAV-10 and EDSV. The data showed that the Taiwan strains could be divided into 2 major groups. The subgroup-1 in group I composes of all Taiwan strains isolated from chickens and 2 Australia strains, CFA3 and CFA40. Whereas mild pathogenic FAV-1A and FAV-10 was grouped in the subgroup-2 in group I. Strains from pigeons and ducks have significant difference from the FAV isolates from chickens and less different from EDSV. Strains isolated from pigeons and ducks was grouped in the same group, group II. The result also indicate that fiber has some relationship in pathogenecity to different animals in addition to being responsible for variations in virulence in the fowl adenoviruses. |
