標題: | 過氧化體增生接受子γ在葛雷夫氏症眼病變扮演角色之研究 | 其他標題: | The study of role of PPARγ in Graves´ ophthalamopathy | 作者: | 黃天祥 | 關鍵字: | 葛瑞夫茲氏眼病變;眼球後纖維母 細胞;過氧化體增生接受子;Graves' Ophthalmopathy;Orbital Fibroblast;Peroxisome Proliferator Activated Receptor γ | 公開日期: | 2000 | 出版社: | 臺北市:國立臺灣大學醫學院內科 | 摘要: | 葛瑞夫茲氏眼病變,簡稱甲狀腺眼病 變,是一種自體免疫性的疾病,病人在眼 球外周圍肌肉組織,眼球後結締及脂肪組 織會大量增生,並且眼球組織浸潤在大量 的免疫性細胞中,主要為CD4+型之T 淋巴 球及吞噬細胞。眼球後纖維母細胞可抑制T 淋巴球的APO-1 蛋白表現量,進而保護T 淋巴球能免於計畫性細胞死亡,使免疫反 應得以持續進行;自葛瑞夫茲氏眼病變的 病人取得之眼球後纖維母細胞,在適當的 荷爾蒙存在下,具有分化成脂肪細胞的能 力。由我們的實驗得知,不罹患葛瑞夫茲 氏眼病變的病人與葛瑞夫茲氏眼病變的病 人之纖維母細胞的過氧化體增生接受子g 形式,其蛋白質含量及與過氧化體增生接 受子核酸序列的結合能力並無不同。但訊 息傳遞活化轉錄因子3 ,在葛瑞夫茲氏眼病 變的病人之纖維母細胞的蛋白質含量,卻明顯比較少。 Graves' Ophthalmopathy (GO), the most frequent extrathyrodial manifestation of Graves' disease, remains a pathogenetic enigma and a therapeutic dilemma. Support for the concept that GO is of autoimmune origin comes from the associated histopathological changes. There is an increased volume of the extraocular muscles and orbital connective and adipose tissues; the extraocular muscles are edematous due to increased production of the hydrophilic glycosaminoglycans (GAGs) in the orbital tissue; a marked infiltration of immunocompetent cells is detectable. Orbital fibroblasts may contribute to perpetuate the ongoing immune reaction in the orbit by protecting infiltrating T cells from apoptosis. Orbital fibroblasts include a subpopulation of cells (preadipocytes), which, under particular hormonal stimulation, differentiate into adipocytes and may contribute to the increased volume of retroorbital adipose tissue. In the present study, we succeed to isolate fibroblasts from minced explants of connective tissue removed in the course of transantral orbital decompression surgery for GO. Using Western blotting and Electrophoretic gel mobility shift assays, the protein level and the DNA binding affinity on PPAR response element (PPRE) of nuclear PPARg in the cultured Graves' and normal orbital fibroblasts are not changed significantly. But the protein amount of signal transducer and activator of transcription (STAT3) is decreased in the cultured Graves' orbital fibroblasts than in the normal orbital fibroblasts. |
