Secretion of aspartic proteinases has been determined to be one of the virulence factors of the pathogenic Candida species. Besides, yeast-hypha morphogenesis is coregulated with SAP4-6 gene products. Thus, Sap4-6 proteins are the potential targets for intervention. Our previous study demonstrated variation of fungistatic activity of anti-Sap5 antiserum and control serum against different bloodstream isolates of C. albicans. In this study, in order to analyze the extracellular proteolytic activities of a large number of Candida clinical isolates, we developed a screening system based on a solid medium containing bovine serum albumin as the sole nitrogen source. Using this system, we assessed 78 clinical isolates of Candida collected from patients treated at National Taiwan University Hospital, for the presence of secreted aspartic proteases (Saps). Implications of such differences on pathogenicity will be determined by evaluation of relationship between Sap levels and patient outcome. |
