作者: | 黃實宏 | 關鍵字: | 細胞黏附分子;甲狀腺癌;cadherin;catenins;thyroid carcinoma cell | 公開日期: | 31-七月-1999 | 出版社: | 臺北市:國立臺灣大學醫學院外科 | 摘要: | 正常上皮細胞之間的adherens junction,是靠cadherin 分子互相結合及附著在 其上的actin filaments 細胞骨架,如此才能構成 一個完整而有功能的adherens junction。此構造 與細胞生長,極化及訊息傳遞途徑有密切的關 係。E-cadherin(135kD)是cadherin 家族的一成 員,屬於一種Ca++-dependent 的穿膜醣蛋白, 大約由700-750 個氨基酸所構成。完整的 cadherin 功能,需要靠細胞質內三種蛋白質a- catenin(102kD) 、b-catenin(88kD) 及g-catenin (82kD)的聯繫,才能使cytoskeleton 接上去。此 複合體已被證明與癌症成因及惡性侵犯程度 有密切關係。在本研究中,我們對正常甲狀腺 組織、甲狀腺濾泡性結節、良性甲狀腺濾泡腺 瘤及惡性甲狀腺濾泡癌作免疫螢光染色及免 疫轉漬分析,來檢查E-cadherin 以及a-、b-及 g-catenin 的表現。在正常甲狀腺組織、濾泡性 結節、良性濾泡腺瘤,此4 種分子皆正常分佈 在細胞與細胞連接處,由免疫轉漬分析也可加 以印證。在惡性濾泡癌,其E-cadherin 及a- catenin 則與在正常細胞之分布無異,但在10 個受檢的組織中,有8 例無法染出b-catenin, 而有7 例無法染出g-catenin。這些結果顯示, 分化型甲狀腺癌之生成因與E-cadherin 本身較 無關係,而b-與g-catenin 的存在與否,可能可 以作為癌症診斷的分子標記。 E-cadherin is a member of the cadherin family that plays a major role in epithelial integrity and tumorogenesis. Catenins are a group of cytoplasmic proteins that regulate the intracellular anchorage of cadherin and are required for the linkage between cadherin and the actin cytoskeleton. Loss of E-cadherin contributes to the pathogenesis in tumor invasion and gives a poor prognosis. In order to investigate the adhesion property of intercellular junctions in thyroid tumors, expression of a-, b-, and g-catenin should also be studied. A correlation between these molecular markers and malignancy would be useful as a preoperative diagnostic test for thyroid neoplasms. The expression of E-cadherin, a, b, and g-catenin were studied in normal and neoplastic thyroid tissue by immunofluorescence microscopy and Western blot analysis. In the normal thyroid and in nodular goiter, and follicular adenoma, staining for E-cadherin, a-, b-, and g- catenin was seen mainly at the lateral surface of epithelial cells in the follicle and the presence of these molecules was confirmed by Western blot analysis. Follicular carcinoma tissue stained positive for E-cadherin and a-catenin, but the results of b- and g-catenin immunostaining were highly variable, with b-catenin being absent in most follicular carcinomas (8/10) and g-catenin being absent in some follicular carcinomas (3/10). These results suggest that E-cadherin expression was not reduced during the pathogenesis of differentiated thyroid malignancies. Impairment of the cadherin-catenin complex at the cell junction may contribute to the malignant progression of differentiated thyroid neoplastic tissue. |
