標題: | 台灣北部沿岸海域中海綿附存型和浮游型發光細菌之多樣性 | 作者: | 謝文陽 | 公開日期: | 2005 | 出版社: | 臺北市:國立臺灣大學海洋研究所 | 摘要: | 本研究在台灣北部沿海不同地區採集了海 水和海綿樣本,針對這些樣本所含發光細菌進 行了計數及分離。平板計數結果顯示,有八個 海綿樣本發光細菌之生菌數介於104~105 CFU/g wet wt,其他樣品生菌數大多小於102 CFU/ml 或 CFU/g wet wt。發光細菌和異營細菌生菌數 比值,除三個海綿様本超過9% (9.0%~18.2%), 其他都小於百分之五。 從各類樣本分離出之發光細菌共計41 株,這些 分離株都是屬於需鹽性兼性嫌氣性革蘭氏陰性 桿菌,都可歸類為弧菌科(Vibrionaceae)菌種。 分離株依其在4°C、40°C 與8%NaCl 的增殖情 形,及是否利用甘露醇或蔗糖為唯一碳源特 性、可粗分為六群。經16S rRNA 基因序列分析 比對發現,第一菌群代表株之最相似菌種為 Photobacterium leiognathi (相似度99.4%)。其餘 五群代表株都與Vibrio herveyi 或其近緣種,具 有98%以上序列相似度;它們的表型特性也都 與Vibrio herveyi 吻合。利用HhaI、DdeI、RsaI 和ScaI 四種限制酶於16S rDNA 限制型分析, 可將六群菌株區分為二個16S rDNA 基因型 群, 第一菌群菌株與其最相似菌種 Photobacterium leiognathi 都屬於第一個基因 型,第二至第六菌群菌株與Vibrio herveyi 都屬 於第二個基因型。研究結果顯示,台灣北部沿 岸海水中發光菌的菌種組成不外 Photobacterium leiognathi 和Vibrio herveyi 或其 近緣種。海綿共存發光菌則以Vibrio herveyi 或 其近緣種為主。 Enumeration and isolation of luminous bacteria from seawater and sponges collected from the coastal regions of northern Taiwan were performed in the present study. Plate counts indicated that luminous bacteria in mostly sponge samples (8 out of 13) were in the range between 104 and 105 CFU/g wet wt; others were less than 102 CFU/ml or CFU/g wet wt. Luminous bacteria accounted for 9.0 to 18.2% of heterotrophic bacteria in some sponge samples (3 out of 13), while only less than 5% of heterotrophic bacteria in the other samples were found to be luminous. Forty-one strains of luminous bacteria were isolated. All of them were halophilic, facultatively anaerobic, Gram-negative rods, and could be identified as the family of Vibrionaceae. According to morphological and physiological characteristics, these isolates were divided into six groups. The first group had the highest similarity with Photobacterium leiognathi (99.4%). The other 5 groups had higher than 98% similarity with Vibrio harveyi or its closely related species. It was found that these V. harveyi or its closely related species and P. leiognathi represented the entire luminous bacterial flora in the shallow coastal waters of Taiwan during the sampling period. The sponge-associated luminous bacteria were predominated by Vibrio harveyi or its closely related species. |
