標題: | 利用添加物來控制藥物透膜速率-輸送機制之探討 | 作者: | 王大銘 | 關鍵字: | 藥物控釋;添加物;界面活性劑;攜帶作用;膨潤現象;controlled release;additive;surfactant;synergetic effect;swelling | 公開日期: | 2000 | 出版社: | 臺北市:國立臺灣大學化學工程學系暨研究所 | 摘要: | 本研究分為兩部份,一部份是探討在親水性藥物溶液中(theophylline)添加疏水性藥物(benzocaine)來提高親水性藥物透過疏水性薄膜的速率;另一部份則是在藥物溶劑中添加界面活性劑來改變藥物透膜速率。結果顯示添加benzocaine 可以大幅提高theophylline 在PU 膜及EVAc 膜中的透過速率,而其提升的機制是源自於benzocaine 的透過可攜帶theophylline 透過,此種攜帶作用與藥物間的作用力有十分密切的關係。添加界面活性劑亦可改變藥物的透膜速率,但其主要機制是界面活性劑會改變溶媒對膜材的膨潤度,提高膨潤度的界面活性劑可提高藥物透膜速率,減少膨潤度者則降低速率。 In this work, benzocaine (a hydrophobic drug) was added in the donor cell to enhance the permeation flux of theophylline (a hydrophilic drug) in the EVAc and PU (hydrophobic) membranes. It was observed that the theophylline flux could be greatly enhanced when benzocaine was added in the drug formula. Evidence was obtained showing that the enhancement effect on theophylline flux was originated from the synergetic transport of benzocaine and theophylline. Data were also obtained indicating that the synergetic transport was strongly related to the interaction between the drugs involved. Besides, the addition of surfactants in the drug formula could also change the drug permeation rate. The effect stemmed from that the addition of surfactant could affect the swelling of membranes. Surfactants increasing the degree of swelling would enhance the drug permeation rate. And surfactants decreasing the degree of swelling would have the opposite effect. |
