2014-11-242024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/681846摘要:皮膚光老化是因長時間曝曬於日光所引起的損傷,臨床表現為皮膚粗糙、鬆弛、皺紋形成、色素沈澱,甚至進一步引發腫瘤生成。UVA 能穿透皮膚真皮層,刺激真皮細胞纖維內的 ROS 增加,抑制膠原蛋白生成與促進基質金屬蛋白酶(Matrix metalloproteinases, MMPs) 表現,進而促進膠原蛋白降解。UVB 所造成細胞的損害尤其嚴重,除了刺激黑色素細胞加速分泌黑色素外,可能還會造成 DNA 或蛋白質等分子的損害,甚至發生癌變。啤酒花 (Humulus Lupulus L.) 富含許多酚類物質,亦含特有成分異戊烯基查耳酮與啤酒花酸,先前研究指出啤酒花當中的活性成分具有抗菌、抗發炎、抗氧化、抗癌等功效。故本研究探討啤酒花中活性成分的抗氧化能力,藉由抑制人類纖維母細胞、人類角質細胞內 ROS 含量,而達到減緩皮膚經紫外光照射後,膠原蛋白流失之保護功效,並進一步評估其抑制黑色素生成與黑色素瘤細胞生長之能力。<br> Abstract: Skin aging is influenced by several factors, and solar ultraviolet (UV) radiation is considered as one of the most important causes of skin photoaging, including wrinkling and hyperpigmentation. Wrinkle formation is relevant to the degradation of extracellular matrix (ECM), especially collagen, the major material that provides structural to the skin. UV radiation promotes the generation of oxygen species (ROS) and synthesis of matrix metalloproteinases (MMPs) in dermal fibroblast. It was suggested that MMPs contribute to the breakdown of dermal interstitial collagen and other connective tissue components. Hops have been attracted a great deal of attention as sources of small molecules such as humulones, lupulones and xanthohumol with the potential for beneficial effects on human health. In this study, we investigate the anti-photoaging of hops extract by suppressing ROS level and MMPs activity in UV-induced human skin cells.
