牙醫專業學院: 臨床牙醫學研究所指導教授: 陳敏慧施聿鎧Shih, Yu-KaiYu-KaiShih2017-03-062018-07-092017-03-062018-07-092015http://ntur.lib.ntu.edu.tw//handle/246246/277056背景:過去研究發現放射線療法,會使活性氧類上升,氧化壓力增加,使唾液腺功能低下。亦有研究發現自體免疫疾病如Sjogren’s syndrome,病患唾液中8-OHdG等DNA被氧化的產物會增加。因此如何避免氧化壓力造成的傷害是一個相當重要的議題,近年來有許多研究,關於是否能從植物中取得抗氧化的物質,來平衡氧化壓力,而金雀異黃素是一種出現在大豆中的大豆異黃酮,具有抗氧化的特性。 因此本研究之假說乃是金雀異黃素有能力保護唾液腺細胞對抗活性氧類(ROS)的傷害。 本研究之目的在於探討探討金雀異黃素是否能夠保護唾液腺細胞,避免活性氧類的氧化,以期能夠進一步提供口乾症患者,改善其症狀的方法。 方法:從一週大的小鼠取其頷下腺唾液腺細胞進行培養。預先加入金雀異黃素處理二十四小時,再放入過氧化氫產生活性氧類進行破壞,並作MTT生物相容性測試、澱粉酶測試,以及免疫螢光染色觀察其型態。 結果:利用MTT生物相容性測試、澱粉酶測試、免疫螢光染色檢視,可發現低濃度(1.25 ⼚3;g/ml)的金雀異黃素在MTT生物相容性測試中有較佳的表現,測試其澱粉酶功能,也有較好的表現,免疫螢光染色的結果中的α-amylase在有金雀異黃素的組別也有較強的染色強度。 結論:本研究證實原本的假說,金雀異黃素是一優良的抗氧化物質,在低濃度下能夠有效保護唾液腺細胞,降低唾液腺細胞受到活性氧類的氧化壓力破壞。Back ground:There are so many reasons cause xerostomia like radiation therapy duo to head neck cancer or autoimmunity disease such as Sjogren’s syndrome. Radiation therapy will cause the rise of R.O.S. resulting the hypofunction of salivary gland. Other research was also noted the rise of 8-OHdG a DNA oxidation maker in saliva of the the patient with Sjogren’s syndrome. It’s an importrnt issue to prevent the oxidation damage. Recently there are so many research about if there are any substrates can act as antioxidant in the plants. Genistein is one kind of isoflavones, and can act as antioxidant. We hypothesized that genistein one of isoflavones can protect salivary gland cells from ROS attack. Specific aim : investigate whether genistein can protect the salivary acinar cell, reduce the oxidation damage, in order to provide a possible treatment for patients with xerostomia. Method: salivary gland cells were acquired from 1-week-old mice, after 3~10 passages cells were pretreated with genistein for 24 hours, then treated with hydrogen peroxide for 0~48 hours. MTT assay, alpha-amylase assay and Immunofluorescence were used to evaluation the result. Result: Accroding to MTT assay, alpha-amylase assay, and Immunofluorescence evaluation. It looks like that with low dosage of genistein (1.25⼚3;g/ml), it can protect the acinar cells, reduce the damage from R.O.S.
