國立臺灣大學農藝學系暨研究所劉麗飛2006-07-262018-07-112006-07-262018-07-112003-07-31http://ntur.lib.ntu.edu.tw//handle/246246/19836本計畫是轉殖TPS(海藻糖合成? ) 基因於水稻優良品種台6農7 號 (TNG67)或台梗9 號(TK9),並在後代中 選育出對乾旱有較強忍受力的水稻。計 畫執行至目前已獲得三種方式調控的 轉殖水稻,這些轉殖水稻大部分為1~ 2 個拷貝,並且可轉錄成mRNA。TPS 蛋白可在體外大量表現,經純化後製成 抗體可進行西方墨點分析。轉殖水稻在 逆境下的表現,初步瞭解可耐鹽,目前 在進行乾旱測試,希望在利用已建立的 耐旱篩選方法中,能獲得耐性較強的植 株。In this project, the TPS gene will be transferred to rice (TNG 67 or TK 9) by Agro-infection methods. We will select the high tolerance of draught in transgenic progeny. Now we obtained transgenic plants that constitutive or stress-inducible promoters drove the TPS gene. The transgenic lines contained a low copy number of transgene that achieved transcription by northern-bolt. The TPS protein was overexpressin in E.coli., therefore will proceed western blot after TPS protein being purified. Our data show the transgenic rice are salt tolerant.
