2007-03-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/698322摘要:本計畫之目的在於建立一適當的理論模型來預測生物細胞之電泳遷移率,並同時進行生物細胞電泳實驗之觀測。將理論結果與實驗結果詳細地分析與比較之後,我們可以估計可解離官能基之表面密度、解離常數、二價陽離子與薄膜內已解離官能基之結合常數等重要參數。本計畫的第一個階段將討論可調整電荷模型之細胞電泳理論。第二個階段將充分探討緩衝溶液對細胞電泳之影響。第三個階段則將進一步考慮離子大小對細胞電泳模擬之影響。全程中亦將進行實驗觀察以修正或驗證所推導模式之適用性。<br> Abstract: The electrophoretic behaviors of biological cells under typical conditions will be investigated theoretically through solving the electrokinetic equations, a set of coupled, nonlinear differential equations describing the electric, the flow, and the concentration fields. In addition to model building, the electrophoretic mobility of cells will be measured and observed to modify the model derived and to justify its applicability. The influences of the parameters key to the present problem such as the concentration of dissociable functional groups, the corresponding dissociation constants, the concentration of divalent cations, and the sizes of ionic species will be examined. The first phase of the proposed study focused on the basic theory of electrophoresis for the case when the surface of an entity is of charge-regulated nature, which mimics the behavior of biocolloids such as cells. The influences of buffer solution on the electrophoretic behaviors of cells will be investigated in the second phase, followed by a thorough analysis which takes the effect of ionic sizes into account in the last phase of the proposed study.
