2007-01-012024-05-17https://scholars.lib.ntu.edu.tw/handle/123456789/684822摘要:此計畫之目的為探討水稻在非生物性逆境(如鹽害、乾旱及冷害)之反應機制。計畫內容分為兩大部分。第一部份:將繼續與另兩子計畫(黃文理博士及張孟基博士之計畫)採分工合作之方式,探討水稻對環境逆境反應之機制。本實驗室主要負責持續性的針對由T-DNA插入突變庫(由中央研究院余淑美博士團隊所建立)所篩選出之非生物性(乾旱、滲透、高鹽及溫度)逆境相關突變株,以PCR之方法得到T-DNA所標的基因之完整 cDNA或啟動子序列,並分析此些基因於臺農67水稻各組織表現的特性及其在各環境逆境下之表現反應。根據研究結果,進一步探討水稻對不同環境逆境反應機制之相關性。此部分之研究結果將提供給此整合計畫之各個參與實驗室以做為其進一步之基因應用的基礎。第二部分:於本實驗室先前之相關研究中,已篩選出數個高鹽(NaCl)逆境相關之水稻T-DNA插入突變株,並已由RT-PCR方式得到此些目標基因(例如: molybdenum cofactor sulfurase),且由初步之分析已知此些基因之表現明顯的受NaCl所調控。因而在此計畫中,亦將接續探討此些基因受NaCl 調控之訊息傳導途徑,並分析其T-DNA插入突變株之生理、生化變化,藉此以探討水稻對高鹽逆境之調控機制。而此些鹽害逆境相關基因之功能亦將以轉殖株進行分析,並將其應用於改良水稻對鹽害之耐受性。<br> Abstract: The goal of this project is to reveal the molecular mechanisms of abiotic stress (e.g. salt, drought and cold stress) tolerance in rice plants. To achieve this, we will cooperate with the other two laboratories (Dr. Wen-Li Hung and Dr. Men-Chi Chang’s lab.) to identify the genes that contribute to the abiotic stress hypertolerance/ hypersensitivity phenotype. First, the abiotic stress-responsive mutants would be screened to from the T-DNA inserted rice mutant pool generated by Dr. Su-May Yu. Following, my laboratory is in charge of using molecular approaches to isolate and characterize the T-DNA target genes or promoters of the T-DNA inserted stress-responsive mutants, and the gene expressions in various abiotic stress conditions would be analyzed. The results would be provided to other cooperated laboratories for further studies. Moreover, since there are several salt-stress responsive genes such as molybdenum cofactor sulfurase have been isolated and characterized in our previous studies, we will continuously study the salt-stress signal transduction pathway that regulate the salt-responsive genes. In addition, the changes in the physiological and metabolic functions of salt-responsive mutants would be analyzed. The functions of salt-responsive genes would also be elucidated by transgenic plants.
