2004-01-012024-05-15https://scholars.lib.ntu.edu.tw/handle/123456789/663239摘要:生這些效果,則可生產高DHA機能禽肉產品。本研究室已能生產高多不飽和脂 肪酸之單胞菌藻類,可供飼養試驗評估這些製品之應用性,並進一步研究此類產 品對肉雞脂質代謝的影響。首先,利用已建立之適當培養條件,於生物反應器大 量培養單胞菌藻,以生產足量之多不飽和脂肪酸製品供肉雞飼養試驗用。 飼養試驗分為四組,分別添加0、1%或3%生物製品,外加一組含1%DHA油處理 組。3日齡肉雞餵飼控制組飼料,適應兩週後才開始正式實驗。以評估肉雞對此製 品之利用。餵飼試驗飼糧三週後犧牲動物,測定肝臟及肌肉中脂肪酸組成及參與 脂質生合成基因FAS及轉錄因子SREBP1和SREBP2的表現。同時測定血液與組織 中膽固醇與脂質濃度。 添加含DHA菌藻製品於肉雞飼料,應可增進肌肉及肝臟中DHA含量,並能改善家 禽肝臟中脂肪代謝、調控肝臟中參與脂肪生成基因的表現,達到減少血液中三酸 甘油酯及膽固醇的濃度。藉由已建立簡易之產多不飽和脂肪酸單細胞生物的培養 方法,以進一步評估直接利用這些產品於家禽飼料中,應能降低成本,且提升肉 品價值。 <br> Abstract: Dietary supplementation of DHA oil can reduce the serum cholesterol and lipid contents of pig above 40%. If the effects can influence poultry animals, it is possible to produce high DHA poultry meat products. We have established the optimal cultural conditions of some microalgaes or fungi that can produce high polyunsaturated fatty acid (PUFA), and , can add to diet for feeding test to estimate the practicability of these bio-products in poultry diet. Furthermore, it will also show the effects of these bio-products on the lipid metabolism of poultry. At the beginning of the experiment, we will apply the optimal cultural conditions to mass culture and collect these microorganisms for broiler feeding test. The feeding test includes 4 treatment batches: level of 0, 1, 3% bio-products and 1% DHA oil will be added to the diets for 3 weeks. The 3-day-old broilers will feed the control diet ( bio-products or DHA oil not added ) for 2 weeks before examinating the feeding test. At the end of the experiment, liver and muscle will be collected to determine the fatty acid composition and the expression of lipogenesis genes FAS and transcription factors SREBP1, SREBP2. We will also determine the contents of cholesterol and lipid in blood and tissue. Dietary supplementation of these DHA - enriched bio-products will increase the contents of DHA in broiler muscle and liver. It may also improve the hepatic lipid metabolism, regulate the expression of lipogenesis genes in poultry liver and reduce the contents of serum triglycerides and cholesterol. By the basis of these established cultural methods of some microorganisms that can produce high PUFA, we can further estimate the practicability of these bio-products in poultry diets.
