2016-08-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/660401摘要:組織工程是一門跨領域科學,結合細胞、支架、生長環境等因子,從醫學及工程的角度,實現重建及再生人類組織或器官。因應老年化社會到來,與年齡之骨科疾病及骨缺損,已然成為重要的公共健康問題;細胞治療技術則是建構在組織工程的基礎上,提供骨組織再生醫學相關研究新穎並具潛力之解決方法。本研究提出一種可培養類硬骨組織團塊(bone-like tissues)的技術,由人類成骨細胞(hOBs)或人類間葉幹細胞(hMSCs)、鈣交聯褐藻酸生物支架(Ca-Alginatescaffolds)和自組裝生物反應系統(self-designed bioreactor system)所建構的平台,提供硬骨(bone)自體移植的功能性類硬骨組織團塊。本研究之生物反應器為封閉式迴流系統,提供基礎細胞培養液,避免更換培養液造成汙染,更可以回饋細胞所分泌之訊息因子;鈣交聯褐藻酸生物支架在系統中提供細胞生長環境,輔助成骨細胞及間葉幹細胞增生、分化並維持功能性。就安全性考量,客製化自體移植避免引發免疫排斥之風險,也避免等待適合捐贈者耗費時間及心理煎熬;就便利性考量,自組裝生物反應系統容易架設;就成本考量,系統所需器皿及管路皆為耗材,不會造成經濟上負擔。此培養技術結合安全性、便利性及低成本等三大優勢,期許未來能夠提供病人安全有效並兼顧生活品質的選擇。<br> Abstract: Tissue engineering is an interdisciplinary field that combines the principles ofbiomedical science and engineering with the goal of achieving human tissue regenerationor reconstruction. Due to the elderly people represented a growing share of the worldwidepopulation, age-related orthopedic disorders and bone defects have become a criticalpublic health issue. Cell-based therapy on the basis of tissue engineering provides a noveland potential solution to bone regenerative research; however, to accomplish realfunctional cells or tissues for clinical use is still a tough challenge.This study presented a platform, which were composed of cells (hOBs or hMSCs),Ca-Alginate scaffolds, and self-designed bioreactor system, to form bone-like tissue forbone autogenic transplantation. Since the self-designed bioreactor is a closed-processperfusion system, it not only offers the basal cell culture media for nutrient transportation,but also prevents cell contamination resulting from media replacement. Moreover, thesecreted cytokine and growth factor can work directly and feedback on the cells in thisdynamic system. The Ca-Alginate scaffold in this system supplied as a cell niche for cellingrowth, proliferation, differentiation, and function maintenance.Concern for safety, personalized bone-like tissues allow autogenic transplantationwithout the risk of immune reactions. In addition, the system is easily to assemble andserves as a convenient tool for researchers or medical doctors.
