2006-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/711692摘要:苦瓜 (Momordica charantia L.) 為葫蘆科 (Cucurbitaceae) 一年生蔓性草本植物,是本省重要夏季蔬菜之一,其販售單價高居第一位,栽培地區遍及全省,若能生產熱帶亞洲特產、高品質、耐貯運的苦瓜,將可進軍國外市場,具有發展潛力。苦瓜為雌雄異花同株,雄花與雌花比例在7:1~25:1之間,其花性會受到溫度、日長及外施植物生長調節劑等條件而改變。苦瓜兼具食用、藥用與觀賞價值,若能探討苦瓜性別變化及其分子機制,將有助於釐清苦瓜性別之決定性因子,應用於提高單株雌花表現及結果數。本研究擬利用苦瓜花芽發育過程中肉眼可見的形態變化,配合埋蠟切片觀察內部組織發育狀況,歸納出雄花和雌花發育階段,並以蛋白質二維電泳分析不同發育階段花芽的蛋白質表現狀況,以期了解不同性別花器發育過程中,蛋白質的表現變化。另外,亦以不同生長調節劑處理苦瓜幼苗,進行蛋白質二維電泳分析,進一步探討苦瓜在蛋白質層次上對荷爾蒙的反應,以及蛋白質表現與花性變化之關係。經比對歸納得到之特殊蛋白質,接著擬進行其cDNA及基因的選殖與分析,除透過雜交分析以瞭解此基因於植株各部位及果實不同發育階段之表現情形,進一步釐<br> Abstract: Bitter gourd is one of the typical tropical fruits originated from Taiwan. It has great economic importance in agriculture and its price is higher than other vegetables. The proportion between staminate (male) and pistillate (female) is 7:1 to 25:1. Sex expression of bitter gourd can be regulated by application of various growth regulators such as GA3, auxin, Ethrel, morphactin. Sex expression in bitter gourd is also influenced by environmental factors such as vernalization and day length. This proposal is intended to use two-dimensional protein gel electrophoresis as the analysis method of sex expression-related proteins in bitter gourd. The morphology, length and microscopic structures of flower buds in bitter gourd will be observed to conclude different development stages in male and female flowers for analysis of protein expression. Total proteins from different organs, male and female flowers during development, and tissues after treatment of various hormones will be analyzed by two-dimensional protein gel electrophoresis. Based on the molecular weight, pI value and expression level of the proteins, special spots of sex expression-related proteins will be defined. According to the partial amino acid sequence information of those proteins, oligonucleotides will be degenerated and used as probe to screen the cDNA library of bitter gourd. Those sex expression-related cDNAs isolated will be characterized in sequence and gene expression level. According to the research items, this proposal can be divided into the following three parts: 1. Expression analysis of total proteins in different organs from bitter gourd. 2. Expression analysis of proteins during flower development of bitter gourd. 3.
