2009-07-062024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/658491摘要:本計畫將利用生長試驗、體外培養與生理生化指標評估抗生素取代物對肉雞腸健康狀態之影響,同時釐清瘤胃細菌素對於肉雞腸道發酵表現與菌相變化之相關性,並探討細菌素與抗生素合併使用時,對藥物敏感性的影響。對生長性能與腸道健康之關係,將評估飼料原料組成和抗生素取代物對白肉雞生長性能、免疫能力發育及腸道組織形態之影響。試驗選用泰黴素(tylosin)與六肽黴素(nosipheptide, NHT)兩種抗生素,配合靈芝發酵產物及開發自瘤胃細菌素為抗生素取代物,添加於飼糧中,針對白肉雞生長性能、腸道黏膜形態、血清抗體含量及血液生化指標之表現等方面進行探討。細菌素機制相關探討以飼糧中添加可表現細菌素之酵母菌,透過血液生化分析與菌相分析,釐清細菌素對肉雞腸道之影響,並藉由體外產氣發酵技術進行接種原測定與分析,同時針對家禽常見菌評估與細菌素與抗生素合併使用時,對藥物敏感性的影響。透過雞隻腸胃道體外培養,利用蛋白質體技術找尋具有潛力的分子生物標記,以評估抗生素及其替代物對腸道生理及免疫之影響。上述工作將可建立抗生素及其取代物對白肉雞腸道生理指標之影響。 <br> Abstract: The purposes of this investigation are to evulate the effect ofantibiotic and its replacer on broiler gut health by growth performance, in vitro culture system and gut physiological biomarker. This study is going to compare the effects of antibiotics (NHT and tylosin) and antibiotic alternatives (Ganoderma lucidum and Bacteriocin) on growth performance and immune responses of broilers. The effect of rumen bacteriocin on broiler gut flora fermentation model and the effects of bacteriocin and various antimicrobials in combination also will be tested. Feedstuff and feed additive will be applied to evulate growth performance, immune responses and the morphology of digestic tract. The yeast which expressed bacteriocin will add in boiler diet, blood biochemical assay and gut flora will be determined to evaluate the gut health situation. The in vitro gas production technique will apply on this process to assay the influence of inoculum source on gas production profiles. The effects of bacteriocin and various antimicrobials in combination on the normal (pathogenic) flora of poultry will be test. Through the in vitro enterocyte culture system, the possibility of applicating the mammal biomarkers in the broiler enterocytes will investigate in order to evaluate the effect of antibiotic and antibiotic alternatives on intestinal physiology and immune responses will establish.
