2008-01-012024-05-14https://scholars.lib.ntu.edu.tw/handle/123456789/660168摘要:本計畫目的:以3年時間開發基因轉殖作物毛狀根,以生產生醫重組蛋白質。 重要工作項目:第一年將以綠色螢光蛋白GFP為報導蛋白質對作物毛狀根組織內表 現量與外泌量進行檢討,以比較煙草與黃豆毛狀根表現重組GFP之效率。第二年 則建構可表現塵蟎 (Dermatophagoides pteronyssinus) 一型與二型過敏原融合蛋白 (fusion protein) 之作物毛狀根,及其生產株之篩選。第三年則建立結合生物反應器 液態培養與 Der p 融合重組蛋白質回收工程之研究室規模生產程序。 預期效益: 本研究計畫之技術範圍涵蓋植物基因轉殖、植物組織培養與重組蛋白質 生產與回收等技術,並有明確之產物 (Der p 融合蛋白) 產出,可做為氣喘過敏病人 在臨床上之應用。<br> Abstract: 1.Purpose In this 3-year of study, we will explore the possibility of crop transgenic hairy roots as the host for production of recombinant biophamaceutical proteins. 2.Work items In the 1st year, using gfp as the reporter gene to evaluate the expression and secretion efficiency of GFP in tobacco and soybean hairy roots. Base on the results of 1st year obtained, we will establish and select the clones of crop hairy roots which can express the Der p fusion protein in the 2nd year. In the 3rd year, we will establish the Der p fusion protein`s production process which includes upstream process of bioreactor culture and downstream process (recovery) of Der p fusion protein. 3.Expectancy of this study The platform we develop here involves in the integration of technique of plant gene transformation, plant tissue liquid culture, and production and recovery of recombinant protein.
