2019-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/711760摘要:台灣靈芝產值約17 億元新台幣,為台灣最盛行的健康食品之一。全球靈芝產值約25 億元美金,主要市場集中在中國、韓國、日本等亞洲國家。靈芝學術上之研究主要以赤芝(Ganoderma lucidum) 與松杉靈芝(Ganoderma tusage)為主,紫芝的研究較少。台灣紫芝(Ganoderma formosanum) 為台灣特有種靈芝,其液態發酵菌絲體萃取物在本團隊先前研究中發現具有抑制酪胺酸&#37238;作用之美白功效。Plastic composite support (PCS) 是一種由聚丙烯與營養成分 (主要為農產廢棄物) 擠壓而成之產物。PCS 生物膜反應器可提供微生物生物膜形成所需之基質且能增加微生物代謝物產量。本研究計畫預計以台灣特有種靈芝-紫芝為原料,並以液態醱酵工程的培養方式降低原料成本。並使用PCS 生物膜反應器於台灣紫芝之菌絲體固定化培養,進一步利用膠體層析純化分離來鑑定有效成分的分子結構,最後以反應曲面法最適化活性成分之大量生產條件,作為未來產業化應用之基礎;並以斑馬魚動物實驗平台作為確效之方式,進一步並釐清台灣紫芝的抗黑色素形成機制,本計畫擬分三年完成。<br> Abstract: The annual business value of lingzhi in Taiwan is about 1.7 billion NTD, which is one of the most popular functional foods. The global business size of lingzhi is about 2.5 billion USD and the main countries are China, Korea, Japan and Taiwan. In the previous studies, the main focus of lingzhi are Ganoderma lucidum and Ganoderma tusage. Studies of Ganoderma formosanum are rare. Two decades ago, G. formosanum, an endemic species of Ganoderma in Taiwan, was first isolated. In our previous study, the inhibitory effect of G. formosanum mycelium extracts on tyrosinase, the central regulatory enzyme being responsible for cutaneous pigmentation; however, the other bioactivities of G. formosanum are still unknown. Plastic composite support (PCS) is an extrusion product of a mixture of polypropylene and nutritional substrate (mainly from agricultural wastes). PCS biofilm reactor, a technique base on cell immobilization, can provide substratum for biofilm accumulation and enhance production of polysaccharide. Nevertheless, PCS has never been used for fermentation of Gannoderma. In this study, we plan to use PCS biofilm reactor to do submerged G. formosanum fermentation which aims to reduce the production cost of skin-whitening compound. We will also purify and identify the active compound and use RSM fermentation method to optimize its production process. The zebrafish platform will also be adopted to assure the compound’s activity and its mechanism.
