2007-08-012024-05-18https://scholars.lib.ntu.edu.tw/handle/123456789/705443摘要:本計劃擬以三年時間對細胞在界面上移動之力學問題進行有系統的研究,最終目的為建立起一項可資準確模擬細胞在界面上移動的理論模式,其中包含下列機制:化學導引效應(chemotatic及haptotaxis)、肌動蛋白聚合作用(actin polymerization)、肌凝蛋白收縮應力(contractile stress)、界面間受器結合(integrin and ligand)阻力、外力(如電)效應、及細胞骨骼(cytoskeleton)與原生質的本構行為等。藉由此一模式,我們可了解更詳細的細胞移動行為,及對細胞移動作出適當的操控,以期對生醫科技的發展有所貢獻。在我們的理論模式推導中,我們將由最基本的物理及化學原理出發,對各項機制並輔以關鍵實驗、或採用文獻中可靠的數據。又對於擬研究的細胞移動操控方式,我們將進行詳細的實驗驗證。<br> Abstract: A systematic study of the cell movement on a surface will be carried out in this three-year project. The goal is develop a comprehensive theoretical model for accurate modeling of the cell movement, including the following basic mechanisms: the chemotatic or haptotaxis effect, the actin polymerization, the contractile stress generated by the crawling of the Mosyin II, the drag associated with the integrin dynamics, the external effect (electricity, for example), and the constitutive behavior of the cytoskeleton and cytoplasm. It is expected that we can have a better understanding on the detailed behavior of the cell movement, designing suitable manipulation method on the cell motility, and thus doing contribution to the development of biomedical application using the present model. During our theoretical derivation, we will stick to the first principle of physics and chemistry, supported with key experiments or reliable data in literatures on different mechanisms.
