戴昌鳳臺灣大學:海洋研究所鄭名君Jeng, Ming-ChunMing-ChunJeng2007-11-272018-06-282007-11-272018-06-282006http://ntur.lib.ntu.edu.tw//handle/246246/56587肉質軟珊瑚(Sarcophyton)與葉形軟珊瑚(Lobophytum)為印度太平洋珊瑚礁上常見的兩個軟珊瑚屬。傳統上,屬及種間係以群體外型及骨針型態之變異作為分類依據。然而,在某些情況下,此兩屬間的形態特徵可能模糊不清,導致種類鑑定的困擾,而且此二屬皆具有雙型珊瑚蟲此一共衍徵,是否為單系群值得探討。我們採集了台灣南部及綠島海域的八種肉質及五種葉形軟珊瑚,共六十一株群體體,以粒線體上兩段蛋白質轉錄基因[鹼醯胺腺嘌呤雙核苷酸脫氫酶(ND6)及核酸錯誤修復系統蛋白(MutS)的同源蛋白(msh1),共1379個鹼基對]分別及結合後,探討其分類和親緣關係。本研究共獲得(含外群)62條msh1序列及56條ND6序列,在250個變異位點中,msh1和ND6分別有169和81個,分別以neighbor joining和maximum likelihood建構親緣關係樹,其結果皆顯示肉質與葉形軟珊瑚並非兩單系群,屬間關係為交互參雜之多系群,造成此現象的可能原因包括系群分化不完全、網狀演化、形態與遺傳分化不一致等。Sarcophyton and Lobophytum are two common soft coral genera on Indo-Pacific coral reefs. Traditionally, their taxonomy is based on colony shapes and the variety and morphology of sclerites. However, in some cases the taxanomy between these two genera is vague. In addition, species of the two genera share a synapomorphy, i.e., the dimorphic polyps. Whether they are monophyletic remains to be studied. Samples of Sarcophyton and Lobophytum from southern Taiwan and Ludao were collected. Protein-coding genes [NADH-dehydrogenase subunits 6 (ND6), and mutS homolog (msh1), 1379 bp] were used to study their phylogeny. Among the total 250 variable sites, 169 and 81 were found in msh1 and ND6, respectively. These sequences (n = 62) were analyzed using maximum-likelihood and neighbor-joining methods in MEGA 3.1 (PC version). Phylogenetic trees reconstructed by both methods revealed a similar pattern suggesting that these two genera are not monophyletic (<5% sequence divergence).
