潘銘正2006-07-262018-07-092006-07-262018-07-092002http://ntur.lib.ntu.edu.tw//handle/246246/28699本計劃的目的為調查鉤端螺旋體在台 灣地區豬及軍犬之流行病學及研究鉤端螺 旋體造成腎小管間質腎炎之機制。利用顯 微凝集反應檢驗659 頭豬血清和127 頭軍 犬血清,並以血清型shermani外膜蛋白萃 取物及重組表現之LipL32 蛋白處理腎小管 細胞。利用RNA 差異展示法及相減雜交法 尋找可能的病原性因子。台灣地區豬的鉤 端螺旋體抗體陽性率達57.7%,軍犬抗體 陽性率為77.2%。外膜蛋白萃取物及LipL32 皆可使鼠腎小管細胞的iNOS 表現、MCP-1 及TNF-a的mRNA 表現量增加。以RNA 差異展示法所得到的prs 基因及相減雜交 法所得到的基因需進一步研究,才能清楚 與致病基因的關係。由本計畫結果得知台 灣地區的鉤端螺旋體在動物中有很高的盛 行率,而外膜蛋白引起腎小管的炎症反應 因子表現量增加。The purpose of the plan is to investigate the epidemiology of leptospirosis in pigs, and military dogs and studies the mechanism of tubulointerstitial nephritis by pathogenic leptospira. A total of 659 pigs and 127 military dogs were examined by using standard microscopic agglutination test (MAT). The pathogenic Leptospira shermani outer membrane extract and recombinant LipL32 protein were administered to cultured mouse tubule cells. We use the RNA differential display technology and subtracted hybridization technology to search the possible pathogenic factors. The seroprevalence of leptospira in Taiwan were 57.7% in pigs and 77.2% in military dogs. We found that outer membrane extract and recombinant LipL32 protein could cause the increase of iNOS, mRNA of MCP-1 and TNF-a of mouse tubule cells. The prospective study of prs gene from the RNA differential display technology and genes from subtracted hybridization technology can help to understand the relationship of the genes and pathogenic mechanism.
